MOTS-c research reference

A 16-amino-acid peptide encoded within the mitochondrial 12S rRNA gene. It is studied as a mitochondrial-derived peptide in cell and rodent models of metabolic signaling and cellular stress responses.

Nomenclature and identity

MOTS-c belongs to a small family of mitochondrial-derived peptides, short open reading frames in the mitochondrial genome that are translated into bioactive peptides. Other members include humanin and the SHLP series. MOTS-c was first reported in 2015 as being encoded within the 12S rRNA region.

The synthetic peptide contains two methionine residues and one tryptophan, which makes it susceptible to oxidation. Mass spectrometry readily shows +16 Da oxidation variants, and these should be considered when interpreting identity data.

What the published literature examines

Reviews describe MOTS-c research in cultured cells and rodent models, focusing on AMPK-related signaling, cellular responses to metabolic and oxidative stress, and reported nuclear translocation under stress conditions. A 2023 review in the Journal of Translational Medicine (Wan et al.) summarizes reported effects and proposed mechanisms related to stress, metabolism, and aging biology. A 2023 review in Frontiers in Endocrinology (Zheng et al.) surveys the same body of preclinical work from a translational-research perspective.

These reviews draw largely on preclinical data. They discuss proposed mechanisms and open questions; they do not establish clinical outcomes.

Analytical and research considerations

With four basic residues (three Arg and one Lys), MOTS-c is strongly cationic, which affects retention in reversed-phase HPLC and can promote adsorption to some plastics and glass surfaces at low concentrations. Oxidized methionine species and deletion sequences are the impurity classes most often seen.

Lot documentation that reports both HPLC purity and a mass-spectrometry identity confirmation is more informative than purity alone, because oxidized variants may co-elute near the main peak.

What a Certificate of Analysis can and cannot establish

A lot COA can

  • Confirm that the tested lot matches the expected identity by an orthogonal method such as mass spectrometry.
  • Report chromatographic purity (typically HPLC area percent) for that specific lot at the stated test date.
  • Tie results to a lot or batch number that can be matched against the vial label.

A lot COA cannot

  • Describe any lot other than the one tested, or material after the stated test date.
  • Establish biological activity, safety, or suitability for any experimental model.
  • Substitute for a researcher's own incoming-material verification where a study design requires it.

Current report status for every catalog item is listed in the Certificate of Analysis library. See also how to read a peptide COA.
